چکیده
ribosome contained twice as many proteins as the bacterial The molecular weight distributions of the ribosomal proribosome. This work was undertaken to compare quantitatively teins from Escherichia coli, Micrococcus lysodeikticus, &othe molecular weight distribution and number of ribosomal proplasmic ribosomes of Euglena and mouse plasmocytoma teins from representative procaryotes and eucaryotes. were compared by electrophoresis in acrylamide gels containing sodium dodecyl sulfate. The gels were quantitatively analyzed by spectrophotometric scanning of the protein bands stained with Coomassie brilliant blue. The weight average and number average molecular weights of the proteins of each ribosomal subunit were calculated. The average molecular weights obtained for the two kinds of bacteria were similar and were significantly smaller than those for the two kinds of eucaryotic proteins. The number of moles of protein per 80 S ribosome was estimated to be between 60 and 80, as compared with 40 to 60 proteins per 70 S ribosome. The increased protein content of eucaryotic ribosomes, as compared with bacterial, is explained both by the presence of a greater number of moles of protein and by proteins of larger average size.

Similarity in the size and number of ribosomal proteins from different prokaryotes.
The ribosomal proteins from nine species of prokaryotes have been compared by polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. The stained gels were scanned spectrophotometrically, the weight and number average molecular weights were calculated, and the detailed distribution of the proteins as a function of molecular weight was determined. By all of these criteria, ...
متن کاملIdentification of a region of Escherichia coli ribosomal protein L2 required for the assembly of L16 into the 50 S ribosomal subunit.
In vitro mutagenesis of rplB was used to generate changes in a conserved region of Escherichia coli ribosomal protein L2 between Gly221 and His231. Mutants were selected by temperature sensitivity using an inducible expression system. A mutant L2 protein with the deletion of Thr222 to Asp228 was readily distinguishable from wild-type L2 by sodium dodecyl sulfate-polyacrylamide gel electrophores...
متن کاملStudies on the Protein Moiety of Plant Ribosomes
The approximate number and molecular weight of pea seedling ribosomal proteins were determined by twodimensional electrophoresis on polyacrylamide gels containing either urea or sodium dodecyl sulfate. The small (40 S) ribosomal subunit contained 32 to 40 proteins and the large (60 S) subunit 44 to 55 proteins. The great majority of ribosomal proteins were basic and had molecular weights betwee...
متن کاملDetermination of somatic and excretory-secretory antigens of Fasciola hepatica and Fasciola gigantica using SDS-PAGE
Human fascioliasis due to unknown species and animal fascioliasis caused by both or one of Fasciola spp. are commonly seen in Iran. To compare electrophoretic patterns of somatic and excretory-secretory antigens of F. hepatica and F. gigantica by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE), the adult flukes were collected from infected slaughtered bovine livers. E/S a...
متن کاملCross-linking study on localization of the binding site for elongation factor 1 alpha on rat liver ribosomes.
Complexes containing rat liver 80 S ribosomes, poly(uridylic acid), phenylalanyl-tRNA, elongation factor 1 alpha, and guanylyl(beta, gamma-methylene)-diphosphonate were prepared. Neighboring proteins in the complexes were cross-linked with the bifunctional reagent 2-iminothiolane. Proteins were extracted and then separated into 26 fractions by chromatography on carboxymethylcellulose. Each prot...
